Clinically validated lead exploration

Biotica’s internal drug discovery focus is on leads with clinical validation for the indications in which Biotica intends to position the optimised drug.

Lead selection
Selecting clinically validated leads offers earlier and more robust proof of concept, since the endpoints for preclinical testing are already established. There are three main types of therapeutic programme based on a clinically validated polyketide lead:

  • Generating analogues of approved polyketides for use in the same indication as the parent but with improved characteristics and IP;
  • Generating analogues of approved polyketides which are optimised for use in indications in which the parent has shown clinical efficacy but has not been approved;
  • Optimizing polyketides with the same target as a different clinically effective compound.

Development strategy
Biotica and its expert advisors construct a development plan focused on generating rapid, robust proof of concept. In Biotica’s nPT-CLN project, for example, the goal is to alter the pharmacokinetic characteristics to reduce systemic side effects while maintaining the immunosuppressive effects via calcineurin binding. The screening cascade is therefore focused on the ADME and PK properties with clear, evidence based success criteria.

Partnering strategy
Biotica develops clinically validated projects to at least preclinical proof of concept, before seeking partners with complementary expertise for further development and commercialization.
